Karl Fischer titration

Karl Fischer titration is a laboratory method for accurately measuring water in a sample. For peptides, this matters because excess moisture can cause degradation, while unaccounted water can make a product's apparent purity look higher than it is.

Karl Fischer titration is a precise analytical chemistry method used to measure exact water content (moisture) in a sample. It relies on a reaction in which water reacts with iodine, sulfur dioxide, a base, and an alcohol in a sealed system. Labs commonly use volumetric titration for higher water content and coulometric titration for trace moisture.

Importance of Karl Fischer Titration in Lab Reports

Karl Fischer titration data in a lab report or Certificate of Analysis (COA) helps you judge true purity and storage stability. Lyophilized (freeze-dried) peptides often retain moisture, so this value is used to calculate water-corrected purity, meaning the peptide amount after water is accounted for.

For example, a peptide reported with 98% purity that also contains 5% water has an actual peptide content of only 93.1%. Moisture also matters for shelf life, because high water content can speed chemical degradation during storage and may compromise product safety and efficacy.

Limitations and Common Misconceptions of Karl Fischer Titration

  • Karl Fischer titration measures water through a chemical reaction, but it does not identify the peptide itself or detect non-volatile impurities.
  • Loss on Drying (LOD) measures total weight loss from evaporating substances, such as solvents or fats, while Karl Fischer titration is specific to water.
  • HPLC purity reports the relative detector signal of the peptide compared with other substances, so it differs from the absolute mass of water measured by Karl Fischer titration.
