Peptide content

Peptide content is the percentage of a sample's total weight that is actual peptide, rather than non-peptide material such as water, salts, and residual solvents.

Also called: Net Peptide Content, NPC, net peptide weight

Peptide content, commonly called Net Peptide Content (NPC), is the share of a freeze-dried peptide powder's gross weight that is actual peptide. Because powders also contain moisture, solvents, counterions, and salts, the peptide content is always lower than the vial's total weight.

How peptide content is determined and used in lab reports

  • Laboratories determine NPC with validated methods such as HPLC assay, Amino Acid Analysis (AAA), or quantitative NMR (qNMR).
  • These methods must be validated for accuracy and precision in accordance with FDA guidelines so results are reliable.
  • NPC matters on lab reports because a vial label usually shows gross weight, not active peptide amount.
  • A vial labeled 5 mg at 70% NPC contains 3.5 mg active peptide; the remaining 1.5 mg is water and salts.
  • For precise solution concentrations, using gross weight alone overstates the actual peptide amount.

Peptide content and peptide purity are different measurements

Peptide content and peptide purity are independent measurements. Purity measures whether the peptide sequence is correct relative to sequence-related peptide impurities. Content measures peptide weight compared with non-peptide materials such as moisture and salts.

A sample can be 98% pure yet only 60% content: 98% of the peptide present has the correct sequence, while the remaining weight of the powder is water, solvents, and counterions. The two metrics describe different aspects of the sample and cannot replace each other.
