Qualification protocol

A qualification protocol is a formal set of tests that confirms a piece of laboratory equipment is installed correctly and functions accurately. It helps ensure that the equipment used to test a sample is reliable before any results are generated.

A qualification protocol is a documented laboratory procedure that proves a specific piece of equipment, such as a mass spectrometer or HPLC instrument, is fit for its intended purpose. This protocol tests the instrument itself rather than the sample being analyzed to confirm that the equipment is suitable for use.

The four standard phases of a qualification protocol

  • Design Qualification (DQ): Checks the equipment design against its intended purpose.
  • Installation Qualification (IQ): Verifies correct delivery and installation.
  • Operational Qualification (OQ): Confirms that the instrument operates as expected.
  • Performance Qualification (PQ): Checks consistent performance under everyday operating conditions, often using criteria such as pump flow rate or detector wavelength accuracy.

The impact of instrument qualification on lab report trust

A qualification protocol is a critical factor when evaluating a lab report, as the reported purity and identity data depend on the proper functioning of the instruments used. If the equipment was never qualified, a claimed '99% purity' could be a misleading value caused by a malfunctioning or uncalibrated machine.

This term is often confused with method validation. The practical distinction is that laboratory instruments are qualified, while analytical methods, meaning the specific tests used on the sample, are validated. A lab report is only reliable if the instrument was properly qualified and the test method was properly validated.
