Solubility testing

Solubility testing measures how much of a substance, such as a peptide, dissolves in a specific liquid. Researchers use it to determine suitable solvents for sample preparation. It does not prove a substance's purity or identity.

Solubility testing measures the maximum concentration a peptide or research chemical can reach in specific solvents, such as water, buffers, or DMSO. This process creates a dissolution profile based on physical properties like amino acid charge and hydrophobicity (water-repelling traits) to determine how the substance behaves in different liquids.

Solubility testing is distinct from purity and identity analysis

When you review a lab report or Certificate of Analysis (CoA), it is important to distinguish solubility testing from purity or identity checks. Solubility data does not prove that a sample is safe, pure, or authentic. A heavily contaminated sample may dissolve well, while a highly pure peptide may dissolve poorly if it is naturally hydrophobic. Quality checks require separate analytical methods, such as HPLC for purity and Mass Spectrometry for identity.

Solubility testing has specific limits and misconceptions

  • Solubility testing measures the maximum amount that can dissolve, whereas dissolution testing measures how quickly a substance dissolves over time.
  • Reconstitution is the physical act of mixing a freeze-dried peptide with a liquid, which is different from the analytical measurement of solubility.
  • This test serves as a practical guide for sample handling to help avoid wasting high-value materials, rather than a standalone quality measure.
