Qualitative analysis

Qualitative analysis is a laboratory test that identifies what is present in a sample. It checks whether the correct peptide or chemical is there, rather than measuring how much of it is present.

Qualitative analysis is a laboratory process that identifies which chemical species are present in a sample without measuring their specific quantities. In the context of peptides and research chemicals, this refers to identity testing, which confirms that the molecule in a vial matches the substance claimed on the label.

Confirmatory methods for peptide identity testing

Laboratories confirm identity by comparing measured properties with a reference standard. The main confirmatory methods include:

  • Mass spectrometry (MS) measures the molecule's exact mass-to-charge ratio to produce a unique molecular fingerprint.
  • HPLC retention time matches the duration a substance takes to travel through a chromatography column against the expected time for a reference standard.
  • Nuclear magnetic resonance (NMR) provides structural detail about the molecule.
  • Peptide mapping confirms the specific amino acid sequence of a peptide.

Identity confirmation must precede purity claims

Qualitative analysis is the critical first step when reviewing a Certificate of Analysis (CoA) before any purity claim carries weight. A CoA may list a purity percentage such as 98% by HPLC, but without identity confirmation, that figure only indicates that 98% of the sample is a single substance - not necessarily the peptide on the label.

Reliable laboratory reports should rely on multiple confirmatory methods because single tests, even advanced methods like mass spectrometry, can produce false positives due to experimental noise. It is important to distinguish qualitative analysis, which answers 'What is it?', from quantitative analysis, which answers 'How much is there?'. A single HPLC chromatogram alone does not constitute a complete identity test.
