Circular Dichroism

CD means Circular Dichroism, a laboratory technique that measures how chiral ("handed") molecules absorb polarized light. The result helps show their 3D folded shape in solution. CD is a structural test; it does not test purity or molecular weight.

Circular Dichroism (CD) is a spectroscopic method used to determine the three-dimensional secondary structure of peptides and proteins in liquid solution. By measuring how a sample absorbs left-handed versus right-handed circularly polarized light, CD spectroscopy creates a structural fingerprint. This fingerprint identifies characteristic folds like alpha-helices, beta-sheets, or random coils, which define the molecule's overall shape.

In laboratory reports, CD analysis is essential for verifying that a synthesized peptide has achieved its correct, functional three-dimensional shape. Because a peptide must be properly folded to remain active, CD directly assesses this state across various conditions. These results ensure that the peptide is not only present but also correctly folded, functional, and structurally stable.

Limitations and distinctions from other testing methods

  • CD is not a direct test for chemical purity and should not be confused with a purity analysis.
  • The results can be distorted by certain impurities or buffer ions that interfere with the light signal.
  • This method differs from HPLC and Mass Spectrometry, which identify substances and weight, and from X-ray crystallography, which requires solid crystals rather than liquid solutions.
